At the Cell and Molecular Biology The Department of Cell and Molecular Biology is one of the most international, broad and distinguished biomolecular departments in Europe, comprising six research programs with about 130 employees.
The linear organization of genes and genetic elements along bacterial chromosomes is not random but follows certain patterns and rules as a result of long-term selection and evolution. Likewise, the linear information is supercoiled and organized into 3D domains and macro-domains to fit the chromosome into the cell, while still facilitating efficient replication, and environmentally responsive gene expression patterns. Horizontal genetic transfer (HGT) of ‘foreign’ DNA into bacterial chromosomes is potentially very disruptive to organization and expression but on evolutionary timescales it occurs frequently and is an important driver of bacterial speciation. Chromosomal HGT is associated with changes in virulence, antibiotic resistance, and clonal success and constraints of HGT are very important biological and biophysical questions that at present are poorly understood.
In this project, you will use new technology to systematically investigate chromosome organization. The technique features a combination of genome engineering, high-resolution imaging and genotyping in a microfluidic platform. You will systematically construct and explore genetic variations to identify the pathways involved in regulating chromosome organization, and study the consequences of HGT on different phenotypes.
You have a passion for science and high ambitions to perform top quality research. You hold a master degree in a relevant subject area, ideally Molecular Biotechnology; you have good knowledge of single molecule tracking of fluorescent molecules in living cells and you are proficient in molecular cloning for bacterial strain construction. You have good programming skills. Personal qualities, ability to work independently and at the same time be able to interact with other researchers is very important. Excellent communication skills in written and spoken English are prerequisites.
Experience with microfluidic techniques for studies of bacterial cells.
