Extraction services at Uppsala Genome Center
The success of any long-read sequencing project will depend mainly on the integrity and purity of the input DNA. At UGC, we have been working with High Molecular Weight (HMW) DNA extractions for many years, and have gained experience with extractions from many different organisms using commercially available HMW DNA extraction kits and traditional methods, as well as in-house developed workflows.
Uppsala Genome Center offers HMW DNA extraction as a service for sequencing projects at (or in collaboration with) SciLifeLab.
We also offer support and guidance to users when planning a sequencing project with us, in order to find the most well suited extraction method for each project.

Extraction methods employed at UGC
Kit-based extractions
NGI-UGC uses kit extraction for samples where major purity issues are not expected or specific protocols have proven successful. We offer extraction using Monarch HMW DNA Extraction Kits or PacBio Nanobind kits.
Phenol-chloroform extraction using SDS or CTAB
Recommended for samples displaying purity issues when using other extraction methods. Especially useful for samples high in polysaccharides, phenols, fats and/or pigments. For this type of extraction, UGC tailors the workflow to each specific sample.

HMW-DNA extractions can be very laborious. We are therefore unable to process a large number of samples simultaneously.
