Aims

The course will provide deepened konwledge of

the relation between structure and function of

proteins as well as practice in genetic change

of the characteristics of proteins. The course contains industry related moments and gives training in planning and performance of an industrial project.

Content

Directed mutagenesis and sequence analysis of DNA.

Construction of a mutated gene in an expression

system. Characterisation of produced gene product.

Cultivation of recombinant bacteria.

Purification, characterisation and activity

analysis of mutated proteins. Modelling of mutant

proteins and design of new proteins with computer

based techniques. Directed mutagenesis applied on a realistic problem.

Laboratory work: Design and production of

modified proteins by molecular graphics and gene

technology methods.

Instruction

Lectures, seminars and laboratory work.

Assessment

Written exam at the end of the course. Practical exercises and laboratory work are assigned 2 points. Resittings are arranged twice each year: at the end of the term and in August.
