Morphological profiling, such as the Cell Painting Assay, holds great potential in digital pathology for studying alterations in cell morphology across different diseases. This study will demonstrate how such an approach, using CellProfiler, can be applied to immunofluorescence images of formalin-fixed, paraffin-embedded human brain tissue affected by either Alzheimer’s disease or Creutzfeldt–Jakob disease, a prion disease. The aim is to analyse the morphology of microglia, the resident immune cells of the brain, and to distinguish between homeostatic, ramified microglia and inflammatory, amoeboid microglia, examining how their distributions vary across different brain regions in the diseased brains.
