The Explore assay
Olink Explore is a proteomics method that provides relative concentrations of detected proteins. The method is based on antibodies and Proximity Extension Assay (PEA). Each protein measured by the method is detected by two antibodies that are conjugated with complementary single-stranded DNA. If both antibodies bind to their protein, the DNA strands can form double-stranded DNA which can be amplified. Using next generation sequencing, the amplified DNA is detected and counted. The sequence data is used to generate NPX values indicating the relative abundance of the included proteins.
SciLifeLab Explore lab
SciLifeLab Explore lab offers analysis with two Olink kits - Explore 3072 and Explore HT.
The technology behind the kits is identical but they provide data from different numbers of proteins. Explore 3072 is modular and analyses from about 300 to over 3000 different proteins from 88 samples. The updated HT method analyses 172 samples in one run and provides data from over 5000 proteins.
Affinity Proteomics also offers Olink Target which is a smaller assay that detects 48 or 96 proteins and is read by quantitative PCR.
