Forskning
Vår forskning är inriktad på att förstå de molekylära mekanismerna bakom Alzheimers sjukdom och andra former av demens. Vi har utvecklat antikroppar som binder specifikt till olika former av Aβ eller α-synuklein, eller andra sjukdomsrelevanta proteiner så som mirkogliaproteinet TREM2. Vissa av antikropparna har även modifierats för att binda till transferrinreceptorn (TfR), som normalt är involverad i järntransport till hjärnan. Interaktionen med TfR fungerar som en molekylär trojansk häst, som transporterar antikroppen in i hjärnan där den kan binda till sitt mål. Denna modifiering ökar antikroppskoncentrationen i hjärnan upp till 100 gånger. De bispecifika antikroppar undersöks både som nya PET-radioligander för diagnostik och som immunterapi
Parallellt med utvecklingen av hjärnpenetrerande antikroppar studerar vi förändringar i blod-hjärnbarriären (BBB) vid neurodegeneration. Vi använder även småmolekylära PET-radioligander för att undersöka patologi i den åldrande hjärnan, med huvudsakligt fokus på proteininlagringar, neuroinflammation och synaptisk dysfunktion.
